and effectively elicit immune response against a wide variety of HIV strains, an

Researchers,
including those from Harvard Medical School in the US, found that the 'mosaic'
vaccine, created by combining pieces of different HIV viruses, is well-tolerated
and generated comparable and robust immune responses against HIV in healthy
adults and rhesus monkeys."
These results represent an important milestone," said
Dan Barouch, the lead author of the study published in The Lancet journal."This
study demonstrates that the mosaic Ad26 prime, Ad26 plus gp140 boost HIV vaccine
candidate induced robust immune responses in humans and monkeys with comparable
magnitude, kinetics, phenotype, and durability and also provided 67 per cent
protection against viral challenge in monkeys," said Barouch.Nearly 37 million
people worldwide are living with HIV/AIDS, with an estimated 1.8 million new
cases every year. A safe and effective preventative vaccine is urgently needed
to curb the HIV pandemic, the researchers said.A key hurdle to HIV vaccine
development has been the lack of direct comparability between clinical trials
and preclinical studies, the they said.To address these methodological issues,
the scientists evaluated the leading mosaic adenovirus serotype 26 (Ad26)-based
HIV-1 vaccine candidates in parallel clinical and pre-clinical studies to
identify the optimal HIV vaccine regimen to advance into clinical efficacy
trials.The trial recruited 393 healthy, HIV-uninfected adults (aged 18-50 years)
from 12 clinics in east Africa, South Africa, Thailand, and the US.
Volunteers
were randomly assigned to receive either one of seven vaccine combinations or a
placebo, and were given four vaccinations over the course of 48 weeks.To
stimulate, or 'prime', an initial immune response, each volunteer received an
intramuscular injection of Ad26.Mos.HIV at the start of the study and again 12
weeks later.
The vaccine containing 'mosaic' HIV Env/Gag/Pol antigens was created
from many HIV strains, delivered using a non-replicating common-cold virus
(Ad26).Results showed that all vaccine regimens tested were capable of
generating anti-HIV immune responses in healthy individuals and were well
tolerated, with similar numbers of local and systemic reactions reported in all
groups, most of which were mild-to-moderate in severity.The researchers also
noted several limitations, including the fact that that the relevance of vaccine
protection in rhesus monkeys to clinical efficacy in humans remains
unclear.
There is no definitive immunological measurement that is known to
predict protection against HIV-1 in humans, they said."These results should be
interpreted cautiously. The challenges in the development of an HIV vaccine are
unprecedented, and the ability to induce HIV-specific immune responses does not

can make test-tube babies, grow human eggs in a lab and reproduce mice from
frozen testicle tissue, but when it comes to knowing how a man’s sperm can swim
to, find and fertilize an egg, scientists are still floundering.
Enormous
advances in treating infertility in recent decades have helped couples conceive
longed-for offspring they previously would not have had. Yet this progress has
also been a work-around for a major part of the problem: Sperm counts are
falling drastically worldwide - and have been doing so for decades – and
scientists say their honest answer to why is: "We don’t knowâ€.
Infertility is a
significant global health problem, with specialists estimating that as many as
one in six couples worldwide are affected. In more than half of those cases,
experts say, the underlying problem is in the male.Most of the focus of
infertility research has been on women, however: on what can reduce their
fertility and on how that can be averted, compensated for or corrected with
treatment. While this approach has produced results - and babies - it has also
left male infertility scientifically sidelined.
Treatments such as in-vitro
fertilization (IVF) and intracytoplasmic sperm injection (ICSI), where the sperm

dropped by more than 50 percent in less than 40 years, according to pooled
research published last year, described by one of its authors as an "urgent

address the basic unanswered clinical and scientific questions in andrology -
the study of male reproductive health - would require research ranging from
large, ideally international, epidemiological studies to detailed lab work to
decipher exactly how sperm cells function. Allan Pacey, a senior lecturer in
andrology at Britain’s Sheffield University, said even at a basic level - the
diagnosis of a possible male fertility problem - the science is lagging.The
techniques of sperm analysis - examining ejaculate under a microscope, counting
the sperm, assessing how well they swim, and seeing what they look like - were
invented in the 1950s, he said.
And we are still doing the same thing now.†So
while estimates suggest as many 1 in 20 young men now have sperm counts low
enough to impair fertility, that remains educated assumption, rather than data
from specific studies. "The quality of evidence we have got in this area falls
way behind that of other branches of medicine,†Pacey said.Attracting funding
for fundamental research into possible environmental impacts on sperm counts -
chemical exposure, for example, or smoking, obesity, or sport and exercise - is
tricky, partly because such studies need vast numbers of people, take many years
and may not give clear answers."In the competitive world of grant funding, there
is a view that male infertility is a problem solved,†Sharpe said.

A woman is at risk of losing
her eye after a botched tattoo job left her crying purple tears, the Metro
reported.Catt Gallinger’s boyfriend decided to give her the tattoo, but it soon
didn't go as she had hoped and is now at risk of going blind.After undergoing
surgery, Gallinger, from Ottawa, Canada, took to Instagram to update her status
on her condition.†If this doesn’t work, my cornea is way too thin and at high
risk of rupturing. If it does rupture before, during or afterwards, I lose my
eye,†she revealed in a post on the social media site.She has also expressed
getting the tattoo was a "massive mistake" as it was so badly done that it looks
like "she was crying purple tears from her sclera tattoo as residue escaped",
the report stated.Gallinger is sharing her story online because she wants to
warn other about the dangers of getting a tattoo.
